Metabolisatiestudie van methyldienolone en methyltrienolone aan de hand van het chimeer muismodel en humane lever microsomen

Dopinggerelateerde producten worden veelvuldig gebruikt in de sportwereld door hun prestatie bevorderend effect en bovendien worden er telkens ook nieuwe producten op de markt gebracht om zo de dopingcontroles te kunnen omzeilen. Hierdoor is het noodzakelijk om methoden te ontwikkelen, zodat detectie van deze producten mogelijk wordt. In dit onderzoek wordt de metabolisatie van twee anabole androgene steroïden, namelijk methyldienolone en methyltrienolone, nagegaan. De metabolieten van beide dopinggerelateerde producten zijn weinig tot niet gekend en zouden op deze manier kunnen opgenomen worden in de routine screening ter detectie. De detectie van de steroïden zelf gebeurt in beperkte mate, want na inname worden ze omgezet in het lichaam tot metabolieten. Deze metabolieten laten een langere detectie van het misbruik toe dan het eigenlijk steroïde. Hierdoor is het dus belangrijk dat het metabolisme gekend is. De voornaamste detectiemethoden die hiervoor worden gebruikt zijn GC-MS en LC-MS.

De studie van de metabolisatie wordt uitgevoerd via in vivo en in vitro technieken. In vitro gaat men een metabolisatiestudie uitvoeren aan de hand van humane lever microsomen, om hierdoor het aantal dierproeven te reduceren en dus de vervanging toe te passen van de 3V’s voor verantwoorde proefdierexperimenten. In vivo maakt men gebruik van het uPA+/+-SCID muismodel die getransplanteerde humane hepatocyten bevatten, waarbij de excretie-urine wordt onderzocht van chimere muizen op zoek naar gevormde humane metabolieten. Er wordt eerst en vooral gebruik gemaakt van niet-chimere muizen om een dosisbepaling uit te voeren en ter controle van de excretie studie, waarna overgeschakeld wordt op het uPA+/+-SCID chimeer muismodel.

Het humaan metabolisme wordt bevestigd door het verkrijgen van dezelfde metabolieten in de humane lever microsomen als in het chimeer muismodel. Via analyse op GC-MS van methyldienolone wordt een metaboliet gedetecteerd die gevormd wordt door de mens en ook wordt bevestigd via detectie op LC-MS2. Bij de analyse van methyltrienolone, die enkel wordt gedetecteerd via LC-MS2, wordt eveneens een metaboliet gedetecteerd die wordt verkregen door het humaan metabolisme. Als metabolisatiereactie vinden hydroxylaties plaats van methyldienolone en methyltrienolone. Hierbij is het belangrijk om in een verder onderzoek de structuur van de metabolieten te achterhalen om methyldienolone en methyltrienolone specifieker te kunnen identificeren in de urine.
