Thermally driven phase transitions in biomolecular systems

Ziektes opsporen via temperatuurmetingen: weg met die koortsthermometer !

Aan de hand van de evolutie van de temperatuur in de tijd kunnen ziektes gedetecteerd worden. De warmte dat wordt toegevoegd aan het lichaam, waardoor de temperatuur verandert, geeft voor gezonde en zieke mensen een verschillend patroon weer. Het vergelijken van beide patronen wijst op de aanwezigheid van een bepaalde ziekte, waardoor verdere stappen kunnen ondernomen worden in de juiste richting om de patiënt te genezen. Het patroon vertelt de patiënt onmiddellijk met welke ziekte hij te kampen heeft. Dit in tegenstelling tot de koortsthermometer, waar enkel de temperatuur van het lichaam wordt weergegeven.

Biologie ontmoet fysica

Het menselijk lichaam functioneert aan de hand van verschillende biologische systemen: vetten, eiwitten en DNA zijn bekende biologische systemen die een belangrijke rol spelen in ons lichaam. Ondanks de negatieve bijklank van vetten in de volksmond, draagt dit biologische systeem bij tot de opbouw van de cel, en staan ze bovendien in voor het leveren van de energie eenmaal de cel is opgebouwd. Naast vetten zijn ook eiwitten belangrijke biologische materialen. Ze staan in voor het opbouwen van de spieren en leveren hun eigen belangrijke functie bij verscheidene biologische processen (om bijvoorbeeld de spieren te doen werken). Tenslotte is onderzoek aan de hand van DNA een hot-topic door de rijke genetische informatie die bevat zit in dit biologische systeem.

Al deze biologische systemen komen bij een bepaalde lichaamstemperatuur in een bepaalde fysische toestand voor (net zoals water onder het vriespunt (0 °C) voorkomt als ijs (vaste toestand) en als waterdamp (gas toestand) wanneer het kookt vanaf 100 °C). Wanneer de temperatuur van het lichaam verandert, en een kritische overgangstemperatuur wordt bereikt, bevinden de systemen zich niet langer in de oorspronkelijke fysische toestand. Bij deze temperatuur wordt een nieuwe toestand bereikt waarbij de geometrische vorm van het systeem verandert. Wanneer de vorm van deze systemen veranderen, worden ook de functies hiervan aangetast. In het menselijke lichaam treedt hierdoor een mutatie op, waardoor de werking van het lichaam wordt aangetast, en de patiënt niet langer als gezond wordt beschouwd.

Lichaam scannen levert resultaat

De techniek bestaat erin om de temperatuur van het lichaam te doen toenemen door warmte toe te voegen. Wanneer het lichaam een bepaalde temperatuur bereikt, zal deze warmte niet langer worden gebruikt om de temperatuur van het lichaam te doen stijgen. De warmte wordt nu volledig gebruikt om het bijhorende biologische systeem van fysische toestand te doen veranderen (net zoals water dat doet bij 0 °C en bij 100 °C). Bij deze welbepaalde temperatuur verandert de functie van het biologische systeem. Door de evolutie van de warmte weer te geven in functie van de temperatuur, en deze te vergelijken voor zowel een gezond persoon, als ook voor de persoon die lijdt aan een bepaalde ziekte, wordt de aanwijzing van de bijbehorende ziekte gegeven. Gebruikmakend van deze techniek kunnen verscheidene ziektes, zoals de ziekte van Lyme, Reumatoïde artritis (een ontsteking van het slijmvlies in de gewrichten), Systematische Lupus en kanker, reeds in een vroegtijdig stadium worden gedetecteerd. De patronen voor de verschillende ziektes zijn uniek en worden aan de hand van een referentie herkent. Eenmaal de ziekte nu opgespoord is, kunnen verdere technieken en behandelingen plaatsvinden om de patiënt te genezen. Door deze temperatuurscan krijgt de geneeskunde op een vlotte manier een idee over de desbetreffende ziekte, zodat de behandeling direct kan starten. Dit in tegenstelling tot bijvoorbeeld de koortsthermometer, die enkel een stijging in temperatuur waarneemt, maar geen enkele aanleiding geeft naar de betreffende ziekte.

Bloedplasma geeft het voorbeeld

Eerder onderzoek op bloedplasma toont de werking van de techniek aan. De techniek werd zowel toegepast op het bloedplasma van verscheidene gezonde personen (afhankelijk van leeftijd, geslacht en afkomst) als op het bloedplasma van patiënten. Het bloedplasma van de mens bestaat uit een complexe samenstelling van biologische systemen. Eén van de belangrijkste componenten hieruit zijn eiwitten, die het lichaam op een juiste manier doen functioneren. De samenstelling van al deze eiwitten zorgt voor een bepaald patroon wanneer een scan wordt uitgevoerd. Elk eiwit ondergaat bij een kritische temperatuur een verandering van fysische toestand (weergegeven door een piek in het patroon), de samenstelling van al deze veranderingen geeft ons een vast patroon. Het vergelijken van het totale patroon van een gezonde en een zieke persoon, levert de directe informatie over de ziekte die dan onmiddellijk behandeld kan worden, zodat de kans op een verdere explosie vermeden wordt.
