Studie naar het genotoxisch potentieel van componenten die kunnen migreren uit bedrukte papieren en kartonnen voedingscontactmaterialen

Hoe veilig zijn papieren en kartonnen voedselcontactmaterialen?

We gebruiken ze dagelijks: papieren en kartonnen voedselcontactmaterialen. Denk maar aan de verpakking van rijst, de doos van een pizza of het papieren zakje van de buurtwinkel. Heeft u zich ooit afgevraagd of die papieren en kartonnen voedingscontactmaterialen wel veilig zijn?

Voedselcontactmaterialen als bron van chemische stoffen

Voedselcontactmaterialen (“food contact materials”, FCM) is een brede term die verschillende materialen omvat die in contact komen met voedsel. FCM zijn samengesteld uit een mengsel van chemische stoffen. In principe is elke chemische stof mogelijk toxisch voor de mens. Dat inherente vermogen van een chemische stof om ongewenste effecten bij de mens te induceren wordt ook de ‘hazard’ van een chemische stof genoemd. Het risico daarentegen is de kans dat een bepaald ongewenst effect zich zal voordoen in de gegeven omstandigheden. Bijgevolg wordt het risico niet enkel bepaald door de hazard van de chemische stof, maar ook door de blootstelling. Dat betekent dat een chemische stof uit een FCM enkel een risico inhoudt voor de consument indien de stof uit de verpakking naar de voeding migreert.

Papieren en kartonnen voedselcontactmaterialen

Papier en karton hebben een lange en succesvolle geschiedenis van veilig gebruik in de voedingsindustrie. Binnen de toepassingen zijn er gradaties in het contact tussen het papier en karton en de voeding. Er zijn toepassingen waarbij er innig contact is tussen het FCM en de voeding, zoals theezakjes en bakvellen en verpakkingen die rechtstreeks in contact komen met de voeding zoals de zakken voor suiker en dozen voor droge- en diepvriesproducten. Daarnaast heeft het een zeer breed scala van toepassingen in transport en distributie.

Gebrek aan wetgeving voor FCM

Er werd reeds aangetoond dat chemische stoffen vanuit FCM naar de voedingsmiddelen kunnen migreren. Aangezien FCM een mogelijke bron vormen van blootstelling aan chemische stoffen, is er nood aan een wetgeving die de consument beschermt tegen de mogelijke risico’s geassocieerd met het gebruik van FCM. In Europa bestaat er tot op vandaag enkel een algemene verordening [EC (No) 1935/2004] en een specifieke wetgeving voor kunststoffen FCM [(EU) No 10/2011]. Voor alle niet-kunststoffen FCM waaronder FCM vervaardigd uit bedrukt papier en karton, is er nog geen specifieke Europese wetgeving beschikbaar.

Genotoxiciteit

Aangezien er voor bedrukt en papieren FCM geen specifieke wetgeving bestaat, kunnen deze FCM componenten bevatten waarvan de toxiciteit niet of onvoldoende gekend is. PRIMISTRA is een onderzoeksproject binnen het WIV-ISP (Wetenschappelijk Instituut voor Volksgezondheid - Institut Scientifique de Santé Publique) dat als doel heeft een strategie te ontwikkelen om binnen het groot aantal componenten dat kan worden verwerkt in bedrukt papier en karton FCM die componenten te identificeren die een mogelijk risico inhouden voor de gezondheid van de mens. Bij de aanvang van het PRIMISTRA-project werd een databank opgesteld die bestaat uit componenten die specifiek aanwezig kunnen zijn in bedrukte kartonnen en papieren FCM. Verder wordt voor elke component informatie uit de meest recente versies van de Europese resolutie over papier en karton, de Zwitserse verordening [SR 817.023.21] en de Europese verordening over kunststoffen [(EU) No 10/2011] vermeld. Die documenten delen de componenten telkens op in twee lijsten: geëvalueerde componenten en componenten die niet of niet recent zijn geëvalueerd. Het resultaat is een databank van meer dan 5000 componenten, ingedeeld volgens geëvalueerd en niet-geëvalueerd.

De meerderheid van de componenten behoort tot de laatste groep en werd niet (recent) geëvalueerd (>80%). Voor die componenten dient te worden onderzocht of ze al dan niet een gevaar inhouden voor de volksgezondheid.

Het is onmogelijk om het volledig toxicologisch profiel voor deze duizenden componenten op korte termijn te karakteriseren. Daarom wordt er eerst gekeken naar het genotoxisch potentieel van de componenten. Genotoxiciteit omvat alle processen die een wijziging veroorzaken in de structuur, informatie inhoud of segregatie van het DNA. Het is een belangrijk toxicologische eindpunt aangezien het geassocieerd is met ernstige effecten op de gezondheid waaronder kanker, spontane abortus en genetische afwijkingen. Bovendien kunnen genotoxische effecten optreden na éénmalige blootstelling aan een lage concentratie van een stof.

Pilootstudie

Uit de databank van het PRIMISTRA-project werden 14 testcomponenten geselecteerd waarvan in de literatuur werd aangetoond dat ze mogelijk genotoxisch zijn. De genotoxiciteit werd in de pilootstudie bepaald met een stapsgewijze methodologie. Het genotoxisch potentieel werd eerst geverifieerd in twee in silico-programma’s, namelijk ToxTree en Derek Nexus™. In silico betekent dat de testen worden uitgevoerd met behulp van een computerprogramma. De programma's zijn beide gebaseerd op hetzelfde principe: componenten met een vergelijkbare structuur vertonen gelijkaardige biologische reacties. De programma’s zijn gebaseerd op de structuur-activiteitsrelatie (SAR), waarbij de biologische activiteit van chemische stoffen wordt voorspeld aan de hand van hun chemische structuur. Deze activiteit kan gelijk zijn aan die van de oorspronkelijke verbinding, maar verschillend in sterkte en ongewenste neveneffecten.

Vervolgens werd aan de hand van gegevens van het Europees Agentschap voor Chemicaliën (ECHA) nagegaan of er resultaten van eerdere in vitro en in vivo genotoxiciteitstesten beschikbaar zijn. Daarna werd de genotoxiciteit in vitro getest met de VITOTOX®-test. De VITOTOX®-test is een bacteriële genotoxiciteitstest gebaseerd op bioluminiscentie waarbij de hoeveelheid licht uitgezonden door de bacteriën een maat is voor de genotoxiciteit.

Op basis van de voorspellingen met de in silico programma’s, de gegevens beschikbaar op de ECHA website en de resultaten van de VITOTOX®-test werd besloten dat 4,4’-diamino-difenylmethaan en 4,4’-oxydianiline genotoxisch zijn. Furfural en 5-aminosalicylzuur worden als niet-genotoxisch beschouwd. Voor de andere componenten zijn verdere testen of literatuuronderzoek noodzakelijk (1,10-fenantroline, ethyl-tolueen-4-sulfonaat, soedaan IV, soedaan III, 1-(methylamino)antrachinon, 7-diethylamino-4-methylcoumarin, 2-chlooraceetamide, 4-nitrotolueen, 2-nitrotolueen en 2-methyl-4-isothiazoline-3-on). Zoals eerder vermeld is de blootstelling een belangrijke factor om mogelijke risico’s in te schatten. Daarom moet voor elke component nagegaan  worden of deze migreert van de FCM naar de voeding en effectief een risico vormt voor de consument.

De 14 componenten die getest zijn vormen maar een klein geheel binnen de databank van meer dan 5000 componenten. Er zijn dus mogelijk nog veel meer stoffen die een genotoxisch effect kunnen uitoefenen en verder onderzoek is bijgevolg noodzakelijk.

Conclusie

Het gebrek aan toxicologische gegevens zorgt ervoor dat de risico’s van papieren en kartonnen FCM tot op vandaag niet gekend zijn.
