Ontwikkelingsstrategieën voor thermofiele nitrificatie ter behandeling van warm afvalwater

Waterzuiverende biotechnologie: Some like it hot

Een ideale zomerdag heeft voor vele mensen een temperatuur van 25 tot 30 °C. In de microbiële wereld zijn er echter gemeenschappen die houden van 45 °C en meer. Deze micro-organismen worden onder meer gevonden in natuurlijke warmwaterbronnen of composthopen. Door gelijkaardige processen in deze natuurlijke omgevingen en in waterzuiveringsinstallaties, kunnen deze thermofiele micro-organismen ook voor de laatstgenoemde toepassing gebruikt worden. Maar ondanks talrijke industriële en economische voordelen, wordt in de praktijk niet bij hoge temperaturen gewerkt. In mijn onderzoek lag de focus op het nitrificatieproces en de mogelijkheid om te opereren bij hogere temperaturen.

Nitrificatie is een conventioneel proces om ammonium (NH4+) via het tussenproduct nitriet (NO2-) om te zetten in nitraat (NO3-), wat vervolgens via denitrificatie omgezet wordt in de niet-reactieve vorm stikstofgas (N2) en dit wordt geloosd in de atmosfeer (Figuur 1). Nitrificatie wordt opgesplitst in 2 deelprocessen, nitritatie (conversie van NH4+ tot NO2-) en nitratatie (conversie van NO2- tot NO3-), wat door verschillende micro-organismen uitgevoerd wordt. Stikstofverwijdering is een standaardproces in waterzuiveringsinstallaties en door de schadelijkheid van reactief stikstof, gepaard met de grote antropogene bijdrage in productie van ammoniak, is dit proces cruciaal. Zowel mens als natuur ervaren negatieve gevolgen bij significante aanwezigheden van ammonium, nitriet of nitraat, zoals vissterfte, eutrofiëring of blauwebabysyndroom. Om dit te voorkomen werden strenge normen voor gezuiverd afvalwater ingevoerd.

Wegens praktische limitatie wordt waterzuivering meestal uitgevoerd op mesofiele temperaturen (15-35 °C). Sommige stikstofbevattende afvalstromen hebben echter hogere temperaturen (> 40 °C), waardoor koelingsenergie nodig is voor de behandeling van dit water. Voorbeelden zijn thermofiele digestaten, industriële afvalstromen (brouwerijen, staalindustrie, mestindustrie…) en huishoudelijk afvalwater in gebieden met hoge seizoensgebonden temperaturen. Implementatie van thermofiele nitrificatie kan, naast de afwezigheid van het koelingsproces, mogelijks resulteren in hogere oxidatiesnelheden (kleinere reactoren mogelijk), lagere slibproductie (kleinere kost voor slibverwerking) en inactivatie van pathogenen. Lagere oplosbaarheid van zuurstof in water bij hogere temperaturen, wordt dan weer gedeeltelijk gecompenseerd door een grotere zuurstofoverdrachtscoëfficiënt. Terwijl denitrificatie reeds aangetoond werd bij 55 °C, blijkt thermofiele nitrificatie een struikelblok te zijn. Door de talrijke voordelen kent dit onderzoeksgebied grote interesse.

Nitrificatie onder thermofiele condities kan bereikt worden via een transitie van mesofiel nitrificerend slib naar hogere temperaturen. Dit onderzoek had 2 grote doelstellingen. Enerzijds werd getracht een zo hoog mogelijke temperatuur te bereiken waarbij nitrificatie uitgevoerd werd. Anderzijds werden 2 reactorsystemen vergeleken: een reactor met gesuspendeerde groei (sequencing batch reactor, SBR) en een reactor met groei van biofilm op carriers (moving bed biofilm reactor, MBBR). Ten gevolge van de hogere robuustheid van een biofilm werd voorspeld dat dit reactorsysteem een temperatuuropdrijving beter kan overleven.

Tot voor kort werd via transitie van mesofiel nitrificerend slib naar hogere temperaturen volledige nitrificatie uitgevoerd bij maximaal 45 °C en hierbij werd een temperatuurstijging van 0.25 °C per dag gehanteerd. In dit onderzoek werd gestart bij een reactortemperatuur van 38 °C, wat na een stabilisatieperiode van 78 dagen opgedreven werd met 0.16 °C per dag tot 40 °C, aangezien geen problemen verwacht werden in dit temperatuurinterval. Vervolgens werd de temperatuurstijging gehalveerd. Deze tragere strategie was enorm succesvol aangezien in de MBBR volledige nitrificatie bereikt werd bij 45.5 °C en in de SBR werd zelfs 49 °C bereikt. Het verschil in reactorperformantie kan verklaard worden door de aanwezigheid van een significante verschuiving van de microbiële gemeenschap in de SBR, terwijl dit fenomeen gering was in de MBBR. Door gebruik te maken van gesuspendeerde biomassa en de temperatuur lineair en traag op te drijven, werden de geschikte micro-organismen dus geselecteerd en gestimuleerd.

Gelijktijdig met de temperatuuropdrijving werden batchtesten (kleine, korte reactortesten) uitgevoerd om adaptatiecapaciteit van micro-organismen aan hogere temperaturen in de reactoren op te volgen. Door de activiteit van slib uit de reactoren te bepalen op de reactortemperatuur, 2 graden hoger en 2 graden lager, konden mede voorspellingen gemaakt worden voor mogelijke activiteit bij verdere temperatuurstijgingen. Via deze testen kon een daling in activiteit van NH4+-oxiderende micro-organismen na 44 °C in de MBBR en van NO2--oxiderende bacteriën na 48 °C in de SBR voorspeld worden. Daarnaast was de wijziging in microbiële gemeenschap in de SBR (shift van NH4+-oxiderende bacteriën naar NH4+-oxiderende archaea) ook op te merken in deze testen, door een wijzigende adaptatiecapaciteit aan hogere temperaturen. Deze testen kunnen dus gebruikt worden in toekomstige experimenten om het falen van reactoren te voorspellen en dit trachten te vermijden door de strategie tijdig aan te passen.

In een conventioneel nitrificatie-denitrificatieproces wordt gebruik gemaakt van autotrofe nitrificerende en heterotrofe denitrificerende bacteriën. Door het gebruik van organische stoffen als koolstofbron bij heterotrofe organismen, wordt in de praktijk een snellere groei waargenomen in vergelijking met autotrofe organismen, welke CO2 als koolstofbron gebruiken. De slibproductie van thermofiele nitrificerende micro-organismen blijkt gelijkaardig te zijn aan deze van mesofiele, maar door de standaard tragere groei van deze autotrofen, is de kost voor slibverwerking hierbij altijd gering. Door de reeds bewezen lagere slibproductie van de thermofiele denitrificerende organismen, wordt de slibverwerkingskost, die gemiddeld een derde van het totale operationele kost bedraagt en hierdoor de grootste bijdrage is, toch aanzienlijk gereduceerd. Dit economisch aspect maakt deze operatiestrategie steeds aantrekkelijker. Thermofiele omgevingscondities hoeven zich echter niet te beperken tot de zuivering van thermofiele bronnen want door de beschikbaarheid van restwarmte, kunnen ook mesofiele afvalstromen kosteloos opgewarmd worden en zodoende van de voordelen genieten.

De SBR bleek het beste reactorsysteem om transitie van mesofiel nitrificerend slib naar hogere temperaturen te bewerkstelligen, in vergelijking met de MBBR, en een lineaire, relatief trage, temperatuuropdrijving is de beste strategie, in vergelijking met de literatuur. Bovendien kunnen batchtesten de adaptatiecapaciteit van de micro-organismen weergeven en dus een eventueel falen van reactoren voorspellen. Het mogelijke falen kan dan eventueel vermeden worden door de operatiestrategie aan te passen. Voor zover bekend in literatuur, is de methode, ontwikkeld in dit onderzoek, de eerste succesvolle methode om een mesofiel nitrificatiesysteem om te vormen tot een thermofiele reactor.
